**Subject:** Request for analysis-ready materials to reproduce PX1 susceptibility models

Dear Dr. Conrad and colleagues,

I am preparing a careful, independent reanalysis of the published PIN/LMD ex-vivo susceptibility comparisons in “Emergence and spread of *Plasmodium falciparum* PX1 polymorphisms associated with decreased susceptibility to antimalarials in Uganda” (Nature Medicine, 2026; DOI: 10.1038/s41591-026-04590-5). We have completed an initial unadjusted analysis of the accessible `Source_dataset1` workbook. It shows directionally similar PIN/LMD differences for DHA, lumefantrine, and mefloquine, and no statistically clear RSA difference, but it does not reproduce the published adjusted model or its exact cohort.

To reproduce the published findings accurately, would you be able to point us to or share, subject to your data-use and ethics requirements:

1. The original analysis script, complete model specification, contrasts, and relevant diagnostics for Extended Data Tables 1 and 2.
2. The sample-matched numeric sequencing-depth and allele-support information used for the >=25x and >=90% inclusion criteria, or the corresponding inclusion list.
3. An appropriately de-identified, analysis-ready table (or the approved separate files and join guidance) containing the sample-matched phenotype values and COI, parasitemia, site, collection year, and K13 covariates used in the adjusted models.
4. Clarification of how the published source workbook relates to the 465-sample depth-qualified analysis cohort, including any assay-specific exclusions.
5. Any data-use, redistribution, attribution, or citation requirements for the source data and supplementary materials.

We will not attempt to infer sample joins or reconstruct missing measurements. We are happy to follow any repository, controlled-access, or collaboration process you recommend, and to cite the original work and data appropriately.

Thank you for considering this request and for making the source materials available.

Best regards,
